Semaglutide Reconstitution Calculator
Semaglutide amounts are worked out in fractions of a milligram, which makes the reconstitution conversion less forgiving than it looks. A single misread mark on a U-100 syringe is a rounding error against a 2.5 mg figure and a ten percent error against a 0.25 mg one — and the only thing that changes how many marks an amount spans is the water you chose.

One unit mark is 0.01 mL. At 2.5 mg/mL each mark carries 0.025 mg; at 5 mg/mL it carries 0.05 mg. Halving the water doubles what a single misread mark costs you — that is the entire reason to plan the mix before drawing.

This tool performs unit arithmetic on numbers you enter, and nothing else. It is not medical advice, it does not recommend a dose, and it is not a suggestion that you use any compound. Most of the peptides these calculators get used with are not FDA-approved for the purposes they are marketed for, are sold through a grey market where purity and labelling are not guaranteed, and many are prohibited in tested sport — check yours with the WADA prohibited-status checker. Any decision to use a compound, and any amount, must come from a licensed clinician who knows your history. The FDA-approved semaglutide products are supplied ready to inject and are never reconstituted by the patient — a powder vial you mix yourself is not one of them, and its contents have not been independently verified.

Frequently asked questions
- How do you calculate semaglutide units after reconstitution?
- Concentration first: vial mg divided by bacteriostatic water mL. Then volume: the amount you are working out in mg divided by that concentration. Then units: volume in mL times 100. Example: a 5 mg vial in 2 mL is 2.5 mg/mL; 0.25 mg is 0.1 mL, which is 10 units on a U-100 syringe. The same 0.25 mg is 5 units if the vial was mixed with 1 mL instead.
- How many units is 0.25 mg of semaglutide?
- Only the concentration decides. At 2.5 mg/mL (a 5 mg vial in 2 mL) it is 10 units. At 5 mg/mL (the same vial in 1 mL) it is 5 units. At 1 mg/mL it is 25 units. Any answer that does not name a concentration is meaningless, which is why this tool asks for the vial and the water before it will convert anything.
- Why are sub-milligram amounts harder to get right?
- Because the absolute error stays the same while the relative error grows. Misreading by one unit mark at 2.5 mg/mL is 0.025 mg — trivial next to a 2.5 mg figure, but ten percent of a 0.25 mg one. The lower the amount, the more it pays to choose a water volume that spreads it across more marks rather than fewer, which is what the 'Water to add' mode solves for.
- Do FDA-approved semaglutide products come as a powder?
- No. The approved finished products are supplied as ready-to-inject solutions at a fixed strength and are not mixed by the patient. A lyophilized powder that you reconstitute yourself is not an approved product, regardless of what the label says, and its identity, strength and purity have not been independently verified. This calculator converts units and cannot tell you anything about what is in the vial.
- Does the amount of water change how many draws a semaglutide vial gives?
- No. The vial contains a fixed mass, so draws per vial is vial mg divided by the amount in mg no matter how much water you add. A 5 mg vial worked out at 0.25 mg gives 20 draws at any concentration. Water changes only where each draw sits on the barrel and how much dead space in a syringe costs you in relative terms.
